You could think of it like this, instead of picking up vibrations, or just guitar or other ways of trying to recognize certain parts in a song that have been mentioned before. You could have a program which could pick up notes which are in bass, and notes which are in treble, that's bass and guitar[s(almost)] sorted,
There are programs out there already which can determine the human voice over other things. Drums I’m not entirely sure about but I’m sure picking out drums would be easier than anything else.
If there are two guitars then that would be tricky, BUT if you have the program which took out treble notes then you could POSSIBLY determine the two( the two being lead and rhythm) by which is the loudest, or clearest, say by setting a decibel mark for which the program can only record over or under. That could separate notes quite easily. You would for sure get them all recorded, only problem after that is cutting and pasting parts which are not where they are meant to be. For example,
Say you recorded all you notes and you had two “fret boardsâ€

wow, sorry, ive been up for over 36 hours, so im sorry if alot of that is out of wack, it could work its very hard, professional programming.
the two guitars could be determined using both volume and frequency. once vocals and drums were taken out.

The amount of signal processing you'd need just in order to split the guitars from the other instruments let alone each other is extremely daunting and difficult to implement. Even then it would be very imperfect.
Yeah, it can be done. But applications of this magnitude often have an entire team of progammers and signal processing experts working on the dev team.
At the end of the day the difficulty of actually fretting a song well pales in comparison to writing an application that splits the tracks then analyzes them and writes a midi notechart.
